抄録: The&＃x20;ascidian&＃x20;larval&＃x20;tail&＃x20;contains&＃x20;muscle&＃x20;cells&＃x20;for&＃x20;swimming.&＃x20;Most&＃x20;of&＃x20;these&＃x20;muscle&＃x20;cells&＃x20;differentiate&＃x20;autonomously.&＃x20;The&＃x20;genetic&＃x20;program&＃x20;behind&＃x20;this&＃x20;autonomy&＃x20;has&＃x20;been&＃x20;studied&＃x20;extensively&＃x20;and&＃x20;the&＃x20;genetic&＃x20;cascade&＃x20;from&＃x20;maternal&＃x20;factors&＃x20;to&＃x20;initiation&＃x20;of&＃x20;expression&＃x20;of&＃x20;a&＃x20;muscle&＃x20;structural&＃x20;gene,&＃x20;Myl.c,&＃x20;has&＃x20;been&＃x20;uncovered;&＃x20;Myl.c&＃x20;expression&＃x20;is&＃x20;directed&＃x20;initially&＃x20;by&＃x20;transcription&＃x20;factor&＃x20;Tbx6-r.b&＃x20;at&＃x20;the&＃x20;64-cell&＃x20;stage&＃x20;and&＃x20;then&＃x20;by&＃x20;the&＃x20;combined&＃x20;actions&＃x20;of&＃x20;Tbx6-r.b&＃x20;and&＃x20;Mrf&＃x20;from&＃x20;the&＃x20;gastrula&＃x20;to&＃x20;early&＃x20;tailbud&＃x20;stages.&＃x20;In&＃x20;the&＃x20;present&＃x20;study,&＃x20;we&＃x20;showed&＃x20;that&＃x20;transcription&＃x20;of&＃x20;Myl.c&＃x20;continued&＃x20;in&＃x20;late&＃x20;tailbud&＃x20;embryos&＃x20;and&＃x20;larvae,&＃x20;although&＃x20;a&＃x20;fusion&＃x20;protein&＃x20;of&＃x20;Tbx6-r.b&＃x20;and&＃x20;GFP&＃x20;was&＃x20;hardly&＃x20;detectable&＃x20;in&＃x20;late&＃x20;tailbud&＃x20;embryos.&＃x20;A&＃x20;knockdown&＃x20;experiment,&＃x20;reporter&＃x20;assay,&＃x20;and&＃x20;in&＃x20;vitro&＃x20;binding&＃x20;assay&＃x20;indicated&＃x20;that&＃x20;an&＃x20;essential&＃x20;cis-regulatory&＃x20;element&＃x20;of&＃x20;Myl.c&＃x20;that&＃x20;bound&＃x20;Tbx6-r.b&＃x20;in&＃x20;early&＃x20;embryos&＃x20;bound&＃x20;Tbx15&＃x2F;18&＃x2F;22&＃x20;in&＃x20;late&＃x20;embryos&＃x20;to&＃x20;maintain&＃x20;expression&＃x20;of&＃x20;Myl.c.&＃x20;We&＃x20;also&＃x20;found&＃x20;that&＃x20;Tbx15&＃x2F;18&＃x2F;22&＃x20;was&＃x20;controlled&＃x20;by&＃x20;Mrf,&＃x20;which&＃x20;constitutes&＃x20;a&＃x20;regulatory&＃x20;loop&＃x20;with&＃x20;Tbx6-r.b.&＃x20;Therefore,&＃x20;our&＃x20;data&＃x20;indicated&＃x20;that&＃x20;Tbx15&＃x2F;18&＃x2F;22&＃x20;was&＃x20;activated&＃x20;initially&＃x20;under&＃x20;control&＃x20;of&＃x20;this&＃x20;regulatory&＃x20;loop&＃x20;as&＃x20;in&＃x20;the&＃x20;case&＃x20;of&＃x20;Myl.c,&＃x20;and&＃x20;then&＃x20;Tbx15&＃x2F;18&＃x2F;22&＃x20;maintained&＃x20;the&＃x20;expression&＃x20;of&＃x20;Myl.c&＃x20;after&＃x20;Tbx6-r.b&＃x20;had&＃x20;disappeared.&＃x20;RNA-sequencing&＃x20;of&＃x20;Tbx15&＃x2F;18&＃x2F;22&＃x20;morphant&＃x20;embryos&＃x20;revealed&＃x20;that&＃x20;many&＃x20;muscle&＃x20;structural&＃x20;genes&＃x20;were&＃x20;regulated&＃x20;similarly&＃x20;by&＃x20;Tbx15&＃x2F;18&＃x2F;22.&＃x20;Thus,&＃x20;the&＃x20;present&＃x20;study&＃x20;revealed&＃x20;the&＃x20;mechanisms&＃x20;of&＃x20;maintenance&＃x20;of&＃x20;transcription&＃x20;of&＃x20;muscle&＃x20;structural&＃x20;genes&＃x20;in&＃x20;late&＃x20;embryos&＃x20;in&＃x20;which&＃x20;Tbx15&＃x2F;18&＃x2F;22&＃x20;takes&＃x20;the&＃x20;place&＃x20;of&＃x20;Tbx6-r.b.
